miR-19b modulates chemokine production in IECs via downregulation of SOCS3.

(a), Caco2 cells were transfected with an equal dose of scrambled ncRNA, pre-miR-19b, or anti-miR-19b and then treated with IL-6 (100 ng/mL) for 24 h. MIP-3α expression was analysed by ELISA. The experiment was repeated three times and the result is presented as the mean±SEM (***p = 0.000014, ncRNA vs. pre-miR-19b,***p = 0.000066, ncRNA vs. anti-miR-19b). (b), Caco2 cells transfected with pre-miR-19b and stimulated with IL-6 (100 ng/mL) for 24 h expressed higher levels of MIP-3α compared to cells treated with IL-6 alone; this outcome was reversed by SOCS3 overexpression. The results are presented as the mean±SEM from three independent experiments (***p = 0.000008, ncRNA+control vector vs. pre-miR-19b;***p = 0.000003, pre-miR-19b vs. pre-miR-19b+SOCS3 vector; ns, p = 0.153, pre-miR-19b+SOCS3 vector vs. SOCS3 vector).
