Inhibition of proliferation, migration and invasion by KMO siRNAs in vitro.

(A) Analysis of KMO expression in HCC cell lines by Western blotting. Upper panel Representative Western blots for KMO and β-actin were shown. Lower panel Band intensities of KMO were normalized to β-actin and showed as Relative Intensity to L02 cells. Uncropped full-length blots were showed in the Supplementary Fig. 3. (B) SK-Hep1 and MHCC-97H cells were transfected with no siRNA (Mock), siRNA control (Scramble siRNA) or siRNAs against KMO (KMO siRNA#1 and KMO siRNA#2), respectively. Knockdown efficiency of KMO was verified by Western blotting. Upper panel Representative Western blots for KMO and β-actin were shown. Lower panel Band intensities of KMO were normalized to β-actin and showed as Relative Intensity to Mock cells, respectively. Uncropped full-length blots were showed in the Supplementary Fig. 5. (C) Cell proliferation of SK-Hep1 and MHCC-97H cells transfected with KMO siRNA was observed by CCK8 assay. (D) Cell migration of SK-Hep1 and MHCC-97H cells transfected with KMO siRNA was measured by transwell migration assays. (E) Cell invasion of SK-Hep1 and MHCC-97H cells transfected with KMO siRNA was measured by matrigel invasion assays.
