Suppression of NF-κB signaling pathway by TBBt.
(A) Mice were injected with 2 mg/kg TBBt and total lysate and nuclear extract were prepared from kidney homogenates. Nuclear translocation of p65 and p50 subunits and phosphorylation of cytoplasmic IκBα were analyzed by Western blotting 15 min after reperfusion. The expression level of iNOS in total lysate was analyzed by Western blotting 24 h after reperfusion. (B) Nuclear proteins were prepared and DNA binding of p65 subunit was analyzed using EMSA15 min after reperfusion. *p < 0.05 and **p < 0.01 vs. sham; #p < 0.05 and ##p < 0.01 vs. VEH.
