Apoptotic mechanism of PEG-TRAIL-vcMMAE conjugates.
(a) Competitive binding of TRAIL or PEG-N109C-vcMMAE with DR4-Fc/DR5-Fc to confirm the internalization pattern of conjugates. Receptor concentration refers to the concentration of DR4 or DR5. The results were expressed as mean ± S.D. (n = 3) (b) Inhibition of TRAIL extrinsic apoptotic pathway via the addition of pan-caspase inhibitor Z-VAD-FMN. Early (B4 gate) and late stage (B2 gate) apoptotic tumor cells were designated in green.
