Ectopic overexpression of Sf-miR-31 using miR-31 mimic sequence induced significant cell death in a Bim-Bax dependent manner.
(a) Sf-miR-31 mimic oligos used for overexpression of miR-31 induced apoptosis in Sf9 cells as evident in the morphological analysis 24h after transfection. Arrow heads in the DIC images mark apoptotic bodies. (b) Caspase-3 activation assay also suggests significant induction of cell death (*P < 0.05). Transfection control (TC) was also used to test the sensitivity of Sf9 cells to the transfection reagent. (c) Significant translocation of Bax to mitochondria was evident using immunofluorescence microscopy and (d) western immunoblotting, 18h after transfection with miR-31 mimics (#,*P < 0.02). Purity of cytosolic and mitochondrial fractions was confirmed using anti-VDAC and anti- GAPDH antibodies, respectively. (e) Significant increase was observed in Bim expression 18h after transfection with miR-31 mimics (* P < 0.02).
