The overexpression of nuclear PKM2 suppresses the apoptosis and cell cycle arrest of CRC cells induced by gefitinib.

(A,C) Flow cytometry was used to analyze the annexin V-positive population of HT29 or C2BBel cells. The final concentration of gefitinib was 10 μM or 30 μM. (B,D) Cleaved PARP and caspase-3 were detected with Western blotting in HT29 or C2BBel cells, which were transfected with PKM2-NLS or PKM2 shRNA. (E,G) Flow cytometry was used to detect the proportions of cells in G1, S and G2-M phases. The final concentration of gefitinib was 10 μM or 30 μM. (F,H) Cyclin D1 and p27Kip1 were detected with Western blotting in HT29 or C2BBel cells, which were transfected with PKM2-NLS or PKM2 shRNA.
