From: SENP1 desensitizes hypoxic ovarian cancer cells to cisplatin by up-regulating HIF-1α
SENP1 maintains HIF-1α stability through desumoylation.
HIF-1α was first immunoprecipitated (IP) with HIF-1α antibody from cell nuclei lysates and the precipitates were then immunoblotted (IB) with SUMO-1 antibody. The transfection of si-SENP1 induced increased SUMO1-HIF1α combination and pl-SENP1 caused the opposite effect especially in hypoxia. Treatment with MG132 (50 μM, 3 hours) enhanced those effects.
