TGY rescued the loss of dopaminergic neurons caused by WT-α-synuclein over expression.
WT-α-synuclein was over expressed by the dopaminergic neuron specific driver Ddc-GAL4. Flies were treated with TGY for 30 days. (A) Brains were dissected and immunostained with TH antibody to visualize the dopaminergic neurons. (B) The number of TH positive neurons in dorsal medial region was calculated (Five brains per group of each experiment). (C) The lysate of the brains were subjected to western blotting analysis and (D) the TH levels of each group were compared by densitometry analysis. To verify the histology and western blotting results, (E) the brains of the flies were extracted for LC/MS analysis to determine the dopamine levels. The results were consistent with (F) the data of climbing assay of each group of flies. *p < 0.05, **p < 0.01 and ***p < 0.001 compared with model flies without treatment. Results were obtained from three independent replicates.
