Analysis of PGK1 mRNA poly(A) tail length and effect of the lack of PAN activity on S. cerevisiae tolerance to acetic acid.
(a) Comparison of PGK1 mRNA poly(A) tail length between wild type, Pab1(+), Pab1A60-9 and Pab1Y514F strains. PCR reactions (12.5 μl) were resolved on 3% agarose gel. Wild type, Pab1(+), Pab1A60-9, pan2Δ and pan3Δ BY4741 strains were grown in minimal medium at pH 3.0 (b) in the absence and (c) in the presence of 40 mM acetic acid. The amino acids histidine, leucine and methionine (50 mg/l) were supplemented. AA: acetic acid, M: DNA ladder. The results of one representative experiment of three are shown.
