miR-10a is downregulated by TNF-α and IL-1β in an NF-κB-dependent manner.

(A) RA FLSs (n = 3 samples) were stimulated with TNF-α or IL-1β (10 ng/ml) for 24 h and the miR-10a expression level was analyzed through real-time PCR. U6B was used as the negative control. (B) RA FLSs (n = 3 samples) were treated with TNF-α for the indicated period and the miR-10a expression level was detected via real-time PCR. (C) Cells were treated with IL-1β and miR-10a was detected as shown in (B). (D) Cells were treated with TNF-α either alone or in the presence of inhibitors of NF-κB (Bay 11-7082), JNK (SP600125) or p38MAPK (SB203580) and miR-10a was detected 24 h later. (E) The same method was applied as in (D), except that TNF-α was replaced by IL-1β. (F) TNF-α-stimulated cells were pretreated with CHX and the miR-10a expression level was analyzed 24 h later through real-time PCR. (G) The same method was applied as in (F), except that TNF-α was replaced with IL-1β. *P < 0.05, **P  < 0.01 or ***P < 0.001 compared with the control group.
