Total RNA samples were prepared from hemocytes of naïve larvae or larvae injected with E. coli, S. aureus and S. cerevisiae (A–C), and real-time PCR was performed for MsDorsal, MsRel2 and MsRel2B as described in Fig. 1. Cytoplasmic (Cp) and nuclear (Nu) proteins were also extracted from hemocytes of naïve and E. coli-injected larvae, and proteins were separated on 12% SDS-PAGE. MsDorsal in the protein extracts was detected by Western blot analysis using rabbit polyclonal antibody to recombinant MsDorsal-RHD (D). Arrow indicates MsDorsal, while arrow head indicates a possible MsDorsal dimer or non-specific protein band recognized by anti-MsDorsal antibody.
