(a) Quiescent VSMC were treated with vehicle (DMSO), plumericin (Plum, 3 and 10 μM) or diamide (DIA, 1 mM) for 30 min before total cell lysates were subjected to immunoprecipitation for Stat3 and immunoblotting for protein-linked GSH (GSSX) and Stat3. Representative blots as well as compiled densitometric evaluations are depicted. (n = 3, *p < 0.05 (vs DMSO control); ANOVA, Dunnett). (b) Quiescent VSMC were pretreated with vehicle (DMSO), 3 μM plumericin (Plum) or 5 μM SU6656 (SU, Src inhibitor) for 30 min prior to exposure to serum (10% NBS) for 10 min. Total cell lysates were subjected to immunoblot analysis for phospho-Stat3 (Tyr705), Stat3 and actin. Compiled densitometric data from three independent experiments are shown in the bar graph (n = 3, *p < 0.05 (compared groups are indicated by lines), ANOVA, Dunnett).
