miR-138 is the upstream regulator of FAK signaling in the promotion of osteogenic differentiation induced by ESW.
(a) Quantitative real-time PCR results show the expression of miR-138 in specified conditions. The miR-138 expression was not affected by the pre-treatment of FAK siRNA. (b) Overexpression of miR-138 dramatically abrogated the benefit effect, i.e. osteogenic promotion, from ESW, as indicated by Alizarin red staining. (c) The ESW induced FAK-ERK1/2-RUNX2 activation was significantly inhibited by miR-138 overexpression as shown in the western blotting results. The expression of OIM + miR-control was normalized as 1. All figure panels: representative data from at least three independent experiments using biological replicates (n = 4).
