From: cJun N-terminal kinase (JNK) phosphorylation of serine 36 is critical for p66Shc activation
Low mitochondrial ROS phenotype of JNK1/2−/− MEFs is reversed by overexpression of p66ShcS36E mutant protein.
Mitochondrial ROS were detected by MitoTracker Red CM-H2XRos dye following treatment with HR (hypoxia 90 min, reoxygenation and staining 30 min) (a) or H2O2 (1 mM, 30 min) (b). Overexpression of p66Shc was confirmed by western blotting (c). Each experiment was repeated at least three times. Statistical analysis was performed using ANOVA (**p < 0.01, ***p < 0.001).
