VOA induced caspase-dependent apoptosis in A172 cells and caspase-independent apoptosis in U251 cells.
(A) Cells were pretreated with pan-caspase inhibitor Z-VAD-FMK (25 μM) for 2 h and then treated with VOA (100 μg/ml for A172 cells and 200 μg/ml for U251 cells) for 48 h. The cell viability was determined by SRB assay. Values represent mean ± SD. *p < 0.05. (B) A172 and U251 cells were treated with indicated concentrations of VOA for 48 h. Then, expression of apoptosis-related proteins including Bax, Bcl-2, pro-caspase 3, cleaved caspase 3, 8 and 9 was determined by Western blotting. The blots were a representative of three independent experiments.
