High glucose (15 mM) treatment reduces IL10’s ability induce STAT3 Tyr 705 phosphorylation.

RAW264.7 cells were cultured for six hours in either 5 mM or 15 mM glucose. Duplicate sets of cells were then stimulated with 1 ng/mL IL10. (a) Cell lysates were separated by SDS-PAGE gel and immunoblotted with antibodies to P-STAT3 Y705 or total STAT3 protein. (b) Band intensities were quantified on a Li-Cor Odyssey scanner and P-STAT3/STAT3 protein ratios are shown in the bar graph. Values are mean ± SEM (N = 3). ***P < 0.01, ****P < 0.001 vs. 5 mM glucose condition.
