The catalytic cycle proceeds via a bi-uni-uni-uni-ping-pong mechanism involving acylation of the catalytic cysteine (C269) and hydride transfer to NAD⁺, followed by trans-thioesterification between the enzyme and CoA to produce the Acyl-CoA product.
