Primary murine SMCs were incubated with compounds 1–13 (1 μM) for 1.5 h and then treated with RAGE ligand CML-AGE (10 μg/ml) for 20 minutes. Cells were harvested and total lysates were subjected to Western blotting with antibodies against phospho-ERK and phospho-AKT. Blots were then re-probed with antibodies to total-ERK and total-AKT and relative optical density (ROD) is reported. (a) Quantified levels of phosphorylated/total ERK normalized to total ERK are shown. (b) Quantified levels of phosphorylated/total AKT normalized to total AKT are shown. Assays results shown are representative of three independent experiments. Error bars represent SEM. *indicates p < 0.05 comparing CML-AGE vs. CML-AGE and compound treatment and #indicates p < 0.001 comparing CML-AGE vs. serum free medium (SFM) alone vehicle. Above each Western blot, S = serum-free medium and C = CML-AGE; numbers above the blots indicate the compound number.
