(A) Chemical structure of SIRT1/2 inhibitor Tenovin-6. (B) Uveal melanoma cells were treated with increasing concentrations of Tenovin-6 for 48 h. Western blotting analysis of acetylated p53, total p53 protein, SIRT1 and SIRT2 was performed after probing with the specific antibodies as labeled. (C) Uveal melanoma cells were exposed to escalating concentrations of Tenovin-6 for 48 h, and mRNA levels of p53 and its target genes (p21, Bax, and Puma) were examined by real-time PCR. GAPDH was used as an internal standard.
