(a) Titration series and kinetics of wild-type HIS-S100A10 binding to immobilized GST-CCR10 CT were determined using surface plasmon resonance (SPR). RU = resonance units, Req = response at equilibrium. (b) Binding of HIS-tagged S100A10 wt and the mutants C82S, C82Q and 85 Stop to GST-CCR10CT was analysed by pull-down and immunoblotting of the glutathione sepharose-bound proteins with the antibodies indicated. Full uncropped scans of blots are shown in Supplementary Fig. S5. (c) Comparison of the KD values ± s.e.m. as determined by SPR. Each titration series was repeated multiple times leading to at least six data points per S100A10 derivative and concentration.
