H1299 cells were transfected with siRNA against GSK3β (a), Flag-GSK3β (b), siRNA against hnRNPK (c), or Flag-hnRNPK (d), and treated with TRAIL (20 ng/ml, 8 hours), then subjected to Western blot analysis with the indicated antibodies. (e,f) Analysis of the cell death rate by Flow cytometry. H1299 cells transfected with GSK3β siRNA or hnRNPK siRNA (e), or transfected with Flag-GSK3β or Flag-hnRNPK (f) were treated with TRAIL (20 ng/ml, 8 hours) and analyzed using Annexin V/PI double staining with a FACS Calibure flow cytometer. (g,h) Statistical analyses of the experiments shown in (e,f) respectively. The analyses were performed with the results of three independent replicates for each experiment. *p < 0.05.
