H2O2 formation in isolated rat heart mitochondria incubated with different concentrations of auranofin in the presence or absence of CsA.
Mitochondria (100 μg proteins) were incubated in 100 mM sucrose, 50 mM KCl, 20 mM HEPES/Tris (pH 7.4), 0.5 mM Na,K-phosphate, 5 mM succinate with the indicated concentrations of auranofin and, when present, 5 μM CsA. ROS production was followed using the fluorescent probes Amplex Red (A) or DHR (B) as described in Methods. The values are the means ± SD of five measurements (***p < 0.001 referred to the respective control).
