(b) Absence of ADAM10 in lysates of ADAM10 (A10−/−) and A10−/−/A17−/− cells and absence of ADAM17 in A17−/− and A10−/−/A17−/− cells was confirmed by Western blotting. β -actin served as loading control. (c–e) The indicated HEK293 knock-out cell lines were transiently transfected with wildtype IL-6R. Cells were stimulated 48 h later as described in the legend of Fig. 2 and sIL-6R was quantified via ELISA. In each panel, one representative experiment out of three performed is shown.
