Pooled MIPs were tested against dilutions of WGA DNA from wild-type (a) B. anthracis, (b) Y. pestis, and (c) F. tularensis. All samples were dual indexed and pooled before sequencing on the Illumina MiSeq platform. Sequencing reads were mapped to reference genes and total mapped reads (left graph) plotted versus the total number of genome copies is represented (The scale of the X and Y axes are Log 10). Error bars represent the standard deviation of three replicates. Cutoffs are shown for each gene as dashed lines. The average percentage of mapped reads for each reference gene at each number of genome copies was plotted (right graph) as a stacked bar chart.
