Activation of NMDA receptors by glutamate induces a down-regulation of Kcc2 and Gabrd transcripts.
(A) Application of glutamate (20 μM) to cultured cortical neurons decreased Kcc2 and Gabrd but had no effect on Gabra5. Transcript abundance was determined one day after glutamate treatment. Data were normalized to Tubb3 and displayed as the geomean of ratios ± s.e.m. [glutamate vs. controls, cell culture experiments (n = 6–11), ***p ≤ 0.001]. (B) Blocking the NMDA-receptors with MK-801 attenuated the post-ischemic down-regulation of Gabrd. MK-801 was injected at the onset of reperfusion. Transcript expression was determined 7 days later.
