(A) Schematic diagram of SH2-U-box to target BCR-ABL or BCR-ABL T315I. (B,C) K562 and K562R cells were transiently electroporated with pFLAG-CMV-4 empty vector (CMV), pFLAG-CMV-4-SH2 (SH2) or pFLAG-CMV-4-SH2-U-box (SH2-U-box). BCR-ABL/BCR-ABL T315I protein levels were analyzed by Western blotting, the relative band intensity of BCR-ABL was derived from three independent experiments and presented as a bar graph (n = 3) (B) and their mRNA levels were measured by quantitative real-time PCR (n = 3) (C). **p < 0.01, Student’s t test.
