APN pre-treatment modulated AMPK signaling and the Bcl2 protein family in FSS-cultured rat BMSCs.
BMSCs were exposed to the FSS environment (15 dynes/cm2) with or without adiponectin pretreatment at different concentrations (10, 20 and 30 μg/mL) for 18 h. Cell lysates were collected as described in the Materials and Methods section and subjected to Western blotting analysis with antibodies specific to p-AMPK, AMPK, p-ACC, ACC, Bcl2, Bax and β-actin; β-actin served as the control. The statistics are representative of bands corresponding to the control and treatment groups. The results are expressed as the mean ± SD, n = 6. aaP < 0.05 vs. the control group; bbP < 0.05 vs. the 15 dynes/cm2 FSS-treated group; ccP < 0.05 vs. the 15 dynes/cm2 FSS-treated + 10 μg/mL APN group; ddP < 0.05 vs. the 15 dynes/cm2 FSS-treated + 20 μg/mL APN group.
