Organotypic Human Cardiac Slice Methodology.
Human atrial and ventricular slices were procured from both (a) non-failing donor (D) and end stage failing (F) hearts. (b) 1cm × 1cm samples from either the left ventricle or the atria were prepared for sectioning. (c) Up to 30 sections were made using a vibratome at a cutting rate of 0.02–0.04 mm/s in ice-cold oxygenated modified Tyrode’s solution. (d) Slices were conditioned at room temperature for 20 minutes before use for either (e) electrophysiology optical mapping, and/or culture and viral transduction.
