(a) Top panel shows LPS silver staining and bottom panel shows loading controls with Coomassie blue-stained proteins prior to proteinase K digests. Bacteria were grown for 24 h in LB, BHI and RPMI at 27 °C and 37 °C. Loading was as follows: odd lanes, parental strain JB580v; even lanes, hfq-negative strain SOR17. (b) Fluorescence of parental strain JB580v and its isogenic hfq mutant carrying a translational fusion of phoP with gfp was measured by flow cytometry. Bacteria were grown in LB for 4 h (log.) and 22 h (stat.). Results are the MFI of at least two independent experiments, each with three independent cultures per strain. Significance was calculated with Student’s unpaired t-test (***P ≤ 0.001; *P ≤ 0.05; ns, not significant, P > 0.05).
