HeLa cells (A), HEK293 cells (B), MARC-145 cells (C) and PAMs (D) were treated with the indicated chemicals at a final concentration of 2 ug/mL and then analyzed using RT-qPCR and Western blotting. The error bar represents standard deviation from three independent experiments. The asterisks indicate a significant difference (p < 0.01) compared to mock transfection. Equal whole cell lysates were subjected to western blotting for analysis of SAMHD1 expression. β-actin was used as a loading control. The fold change of SAMHD1 is expressed as densitometric units of the band normalized to the β-actin level, relative to the control. Uncropped images of blots are shown in Supplementary Figure 3.
