(A) SiiF-HaloTag, HilA-HaloTag, SpaS-HaloTag, or FliN-HaloTag were labelled with TMR-Star and imaged as described for Fig. 4. Selected frames from series of 500 frames are shown (frame rate: 15 frames per second) and frame numbers are indicated. Inserts in frame 2 show localized images. Each trajectory has a different colour. Using pooled trajectories for proteins in at least 20 bacteria recorded under the same conditions, the diffusion coefficient D was calculated using the Jaqaman algorithm5,20,61. (B) Values for the diffusion coefficients. Scale bars, 0.5 μm (SiiF-HaloTag), 1 μm (SpaS-HaloTag, FliN-HaloTag, HilA-HaloTag).
