Comparison of the internalization pathways of M32, H32 and Trastuzumab IgGs binding to HER2-ECD on cell surface.
(A) SKBR-3 cells were incubated with trastuzumab, M32 or H32 IgG for 1 hour and the localizations of the internalized IgGs in relation to those of Rab7 were revealed by immunofluorescence microscopy. (B) Same description as in panel (A) for Rab9. (C) Same description as in panel (A) for Rab5. (D) Relative expression levels of Rab7 and Rab9 in the SKBR-3 cells treated with trastuzumab, M32 or H32 IgG were analyzed by Western blot, where GAPDH was detected as internal control. The Western blots were derived under the same experimental conditions from the same cell lysates; the original full-length Western blot images are showed in Supplementary Figure S4. The expression levels were reproducible with three independent experimental repeats.
