(A) Input 125I-PrPSc. (B) Plasma at 3 hr after p.o. administration. (C) Plasma at 24 hr after p.o. administration. Plasma samples were obtained at designated times after p.o. administration of 125I-PrPSc in fasted mice. Radioactivity was normalized by TCA precipitation. The retention times of peaks corresponded to multimeric PrPSc as seen in the input 125I-PrPSc. Radio-HPLC chromatograms were conducted with BioSep-Sec-S4000 column with a guard cartridge, at the flow rate of 1 ml/min of 25 mM sodium phosphate buffer, pH 7.4. Column eluates were collected at 1-min interval for first 5 min, thereafter at 10-sec per fraction. Column recovery of loaded radioactivity was >80%. Radioactivity was measured by gamma counter.
