Figure 3: β1-AA increased cell viability of cardiac fibroblasts (CFs).

Figure 3

(A,B) Different concentrations of β1-AA and β12-AR agonist isoprenaline (ISO) were added to cultured cardiac fibroblasts respectively for 24 h, then the cell viability was detected by Cell Counting Kit-8. n = 6 per group. (C) CFs before different treatments were labeled with 5 μmol/ml CFSE and cell proliferation was measured by flow cytometry. Data shown here are representative of one of three individual experiments with similar results. The bar graph showed the statistical results. (D) The morphology of cardiac fibroblasts treated with β1-AA and ISO. Vimentin, which is a marker of cardiac fibroblasts, was detected using immunofluorescence and observed in confocal microscopy. After β1-AA and ISO were added to cardiac fibroblasts and allowed to incubate for 24 h, the morphology of cardiac fibroblasts changed. The cells had more cytoplasm and there were more two-nucleus cells. All data are presented as the mean ± SEM of three independent experiments. **P < 0.01 vs. vehicle group, ##P < 0.01 vs. IgG group. (β1-AA negative IgG). IgG: β1-AA-negative mouse IgG.
