PDGF-BB and/or IGF1 regulated the differential expression of IGF1R and CXCR4.
(a) In western blot analysis, recombinant IGF1 decreased the expression of IGF1R in hDSCs with a dose-dependent manner. IGF1 treatment in hDSCs increased the expression of CXCR4. (b) PDGF-BB protein dose-dependently upregulated the IGF1R and CXCR4 expression. (c) In addition, recombinant PDGF-BB reversed IGF1-induced downregulation of IGF1R effectively. (d) hDSCs treated with IGF-I and PDGF-BB revealed the same level of Stat3 phosphorylation as the cells treated with PDGF-BB alone. However, even in both growth factors simultaneously adding to cells, IGF-I did not show an additive effect on p-Stat3 activation.
