Differentiation of patient-specific iPSCs into cardiomyocytes.

(a) Beating cardiomyocytes were generated from control-iPSCs and RA-iPSCs after 12 days of cardiac differentiation. (b) The cellular structure and expression of cardiac markers in iPSC-derived cardiomyocytes were confirmed by immunofluorescence staining and confocal microscopy. Control-iPSC-CMs and RA-iPSC-CMs both showed characteristic sarcomeric structures and positive staining with antibodies against SA-actinin, TNNT2 and MLC2a (scale bar, 200 μm). (c) The mRNA level of a cardiac-specific gene, TNNT2, before and after cardiac differentiation in various cells was determined by quantitative reverse transcription-polymerase chain reaction. Data represent mean values determined in three independent experiments ± SEM. **P < 0.01.
