Silencing mTOR gene in DCs increased PD-L1 expression.

(A) Bone marrow DCs were cultured and transfected with MyD88, TRIF and mTOR siRNA as described in Fig. 1. Forty-eight hours after transfection, DCs were collected and stained with PE-labeled PD-L1 antibody. The expression of PD-L1 was detected by flow cytometry. (n = 4 experiments for the data presented, p < 0.05, mTOR siRNA or combined with MyD88 and TRIF siRNA vs scrambled siRNA transfected cells). (B) The total RNA of DC was extracted and relative quantity of PD-L1 mRNA was detected by Real-Time RT-PCR. (n = 4 experiments, *p < 0.05, mTOR siRNA or combined with MyD88 and TRIF siRNA vs scrambled siRNA transfected cells).
