Regeneration of TACE zymogen using TPD.
Schematic representation of TACE inhibition by TPD: TACE is generated as a zymogen form (a) which undergoes proteolytic cleavage of the auto-inhibitory prodomain in two furin sites by furin (b). The activated TACE is then delivered to the cell surface in secretory vesicle (c) to the cell membrane (d) to shedding its substrates such as TNFα. The addition of TPD restore the zymogenic form (e,f) inhibiting specifically its catalytic activity.
