Hypoxia potentiates chemoresistance to cisplatin in OSCC cells.
(A) HSC3, SCC3, and TCA8113 cells were treated with cisplatin under normoxic (20% O2) or hypoxic (1% O2) conditions for 48 h. The 50% inhibitory concentration (IC50) values of cells was analyzed by MTT assay. Absorbance was measured at 570 nm (A570). (B) Expression of HIF-1α mRNA in HSC3, SCC3, and TCA8113 cells transfected with or without HIF-1α siRNA under hypoxic conditions for 48 h. (C) Expression of HIF-1α protein in HSC3, SCC3, and TCA8113 cells transfected with or without HIF-1α siRNA under hypoxic conditions for 48 h.
