Figure 6

Inhibition of platelet aggregation by Exisulind.
(A) Platelet aggregometer with platelet rich-plasma from a healthy donor showed that ADP-induced platelet aggregation was reduced by PKG activation using cGMP. Reduced platelet aggregation by PKG activation was reversed by PKG inhibitor (8-Rp-cPT-cGMP: 20 μM). (B) Western blot for VASP phosphorylation (indicates inhibitory activity of platelet aggregation) demonstrated that Exisulind treatment increased VASP phosphorylation and that PKG inhibitor (8-Rp-cPT-cGMP: 20 μM) reversed this effect. p-VASP239 = phospho-VASP at serine239, t-VASP = total VASP. Exi = Exisulind. (C) Platelet aggregometer with platelet rich-plasma from rat peripheral blood treated with vehicle or Exisulind for 3 days or 2 weeks. *P = 0.02 vs. Control at Day 3, **P = 0.05 vs. Control at Day 14. The data are presented as mean + SEM of five independent experiments.
