Reactivated latent T cell lines are targets of ADCC.
(a) Jurkat-derived CD4+ T cells were infected with VSV-G pseudotyped, CCR5-tropic NL4.3.ADA.IRES.GFP WT virus and sorted for GFP- cells by flow cytometry. Following a 7-day recovery, GFP- cells were treated with TNFα for 24 h and GFP+ cells were sorted and maintained in the absence of TNFα as latent cells. These cells could be reactivated with TNFα or other latency reversing agents. (b-d) Resting latent cells from Panel a (Step 7) were exposed to romidepsin (RMD) or TNFα for 9 h and examined by flow cytometry for reappearance of GFP (b) and for cell-surface CD4 and BST2 expression, as well as Env recognition by PGT121 (c). Reactivated latent cells were assessed for their susceptibility to FACS-based ADCC (d). Shown in Panel d are ADCC activities from 3 donors as examples.
