Infectivity of PKH67 labeled viruses.

(a) Host cell imaging of S. islandicus ΔC1C2 infected with SMV1. Cells were stained with DAPI and SMV1 particles were labeled with PKH67. The co-localization of DAPI stained cells (blue, left panel) with virus particles (green, middle panel) appears as light purple (right panel). Bar = 9 μm. The insets in (a) represent magnified (2x) views of the selected regions. The Pearson’s and Mander’s overlap coefficients are represented as the average of 6 individual images of infected cells at initial stage of virus infection, 5 min post infection (mpi) and 30 mpi, at an MOI of 10 (Panel b). The infectivity was measured by a plaque assay before and after labelling, and presented as PFU/mL (Panel c).
