SiRNA of AKT and PD suppressed the proliferation, induced apoptosis and decreased phosphorylation of 4E-BP1.

After knockdown of AKT, cells were treated with 10 μM PD for 48 h. (A) The cell morphologic changes were observed by In Cell Analyzer 2000 and typical images were presented. Scale bar: 50 μm. (B) The cell viability was evaluated by MTT assay. (C) Apoptotic cells were stained with Annexin V/PI and analyzed by flow cytometry. Results shown were mean ± SD for three independent assays. (D) Cell extracts were analyzed for the levels of EGFR, HER-2, p-AKT, AKT, p-4E-BP1 and 4E-BP1 by western blot analysis.
