Rg1-induced up-regulation of GR and HDAC2 was mediated by the inhibition of ROS production.

(A) HaCaT cells were pre-treated with N-acetyl-L-cysteine (NAC, 2 mM) or Rg1 (50 μM) for 1 hour and subsequently exposed to UVB (60 mJ/cm2). Fluorescence was measured using CellROX reagents. ap < 0.01 vs. the UVB-irradiated group. (B) HaCaT cells were pre-treated with Dex (1 μM) and/or Rg1 (50 μM) for 1 hour and subsequently incubated with H2O2 for 30 minutes. Then, the cells were treated with 10 ng/ml TNF-α for 24 hours. Western blot analysis was performed using anti-GR and anti-HDAC2 antibodies. (C) IL-6 levels were assessed using ELISA. (D) IL-8 levels were assessed using ELISA. At least two independent experiments revealed highly comparable results. aap < 0.01 vs. vehicle. bbp < 0.01 vs. TNF-α only. cp < 0.05, ccp < 0.01 vs. TNF-α+UVB.
