Pull-down experiment using (a) GST-rNedd4-WW2 and GST-rNedd4-WW3/4, (b) GST-hNedd4-WW3* and GST-hNedd4L-WW3* and U24-6A and U24-7 protein. U24-6A and U24-7 are loaded in the first lane for reference, and the GST tag alone plus U24 are in the second lane. Lanes 3 and 5 are loaded with GST-Nedd4-WW domains alone. GST-Nedd4-WW domains plus U24 are loaded in lanes 4 and 6. In each case, the bottom gel shows that the amount of GST-tagged protein used is uniform for each combination tested.
