Human resting naïve, TCM and TEM cells were isolated and activated with anti-CD3 and anti-CD28 beads for 40 min, 150 min or 15 hrs. Expression of the cytokine genes, IFNG (a), IL13 (b) and IL4 (c) was measured by RNA-Seq. Active chromatin marks (H3K4me1, H3K4me3, H3K27ac and H2A.Z) and RNA Polymerase II were profiled using ChIP-Seq in resting cells (0 hours). The Genome Browser tracks of the ChIP-Seq signals for the IFNG (d), and IL4/13 (e) loci are shown. Y-axis shows coverage by estimated fragments normalized to millions of reads mapped (FPM). Putative regulatory elements are denoted on top and those areas that undergo epigenetic changes were framed. Purple arrows show the locations of combined GATA3/MYB elements as described previously34.
