DCs were seeded at 1 × 106 cells/ml in fresh medium and stimulated for 24 h with 10 ng/ml LPS, in the absence or presence of 2.5 mM 2-deoxy-glucose (2-DG). Control cells (Ctl) received PBS only. Glucose consumption and lactate production were monitored using enzymatic detection kits (a,b). Surface expression of DC maturation marker CD86 was monitored by cell flow cytometry (c) and cytokine secretion was assayed in DC supernatants (d,e). Means ± SEM from 3 experiments are shown. *P < 0.05; #P < 0.01 compared to LPS-stimulated cells.
