The inset shows a schematic illustration of the three different 100 bp DNA scaffolds used, each with 12-base-mismatched regions, located at 0, 4 and 8 bases from the nucleoprotein filament assembly site (red strand). Moving the 12-basepair-mismatched region away from the assembly site significantly decreases the yield of nucleoprotein-filament assembly. The data correspond to the average of three measurements, and the error bars show the standard error of the mean.
