(A) The PCR analysis of regenerated alfalfa plants, the positive control (pBI121-GoGID1 plasmid), the negative control (wild-type), and empty vector control (CTRL). The sizes of the DNA fragments were 795 bp for NPTII (upper), 1812 bp for GUS (middle), 553 bp for part of the 35S-GUS sequence, and 1585 bp for the GoGID1 gene plus 553 bp. (B) The transcript abundance of GoGID1 in transgenic plants was measured by quantitative RT-PCR. The alfalfa actin gene was used as a reference for normalization. (C) Southern-blot analysis of HindIII-digested genomic DNA from leaves of the wild-type and GoGID1-over-expressing lines. The DNA was probed with the 795-bp NPTII fragment from the pBI121 vector.
