3-dimensional MDA-MB-231 cell spheroids were generated in low-adhesion round bottom 96 well plates then embedded into type-I fibrillar collagen derived from rat-tails (450 μg/mL). Collagen-embedded spheroids were then treated with dexamethasone (100 nM) or vehicle control (0.01% DMSO) for 30 min prior to the addition of fetal calf serum (FCS, 5% v/v). Static images were taken every 24 hours for 96 h total using an Olympus IX51 microscope (A). The cell-containing area was traced using ImageJ software to determine the extent of invasion (B). Data are expressed as the area of invasion in pixels/1000 and presented as mean ± SEM from n = 8 independent experiments. *P < 0.05, ***P < 0.001 from two-way ANOVA with Bonferroni post-hoc test.
